    The effect of manipulating training demands and consequences on experiences of pressure in elite netball

    Testing the efficacy of a pressure training framework (Stoker, Lindsay, Butt, Bawden, & Maynard, 2016), the present study investigated whether manipulating training demands and consequences altered experiences of pressure. Elite Netballers (Mage = 26.14 years) performed a Netball exercise in a randomized, within subject design with four conditions: a control, consequences, demands, and demands plus consequences condition. Compared with the control, self-reported pressure was significantly higher in the consequences and demands plus consequences condition, but not in the demands condition. The findings provide mixed support for manipulating demands and strong support for manipulating consequences as a means for producing pressure.     A Full-Ocean-Depth Rated Modular Lander and Pressure-Retaining Sampler Capable of Collecting Hadal-Endemic Microbes Under \u3ci\u3ein situ\u3c/i\u3e Conditions

    The hadal zone remains one of the least studied environments because of its inaccessibility, in part because of hydrostatic pressures extending to 110 MPa. Few instruments are capable of sampling from such great depths. We have developed a full-ocean-depth-capable lander that can be fit with sampling packages for the collection of still images, video, motile megafauna, and hadal seawater. One payload includes a pressure-retaining sampler (PRS) able to maintain seawater samples under in situ pressure during recovery. We describe the technical specifications of the lander and the PRS and preliminary results from three deployments at depths in excess of 10,700 m in the Mariana Trench. Seawater from full-ocean depth was recovered at 81% of the in situ pressure. This facilitated the collection of microbial genomes affiliated with the family Flavobacteriaceae within the Bacteroidetes and the phylum Marinimicrobia. We show that these microbes are specifically enriched in hadal zones, representing novel trench lineages, and describe their adaptations for living in hadal environments. These findings highlight the utility of this lander system, which facilitates scientific inquiry at depths greater than 6000 m

    Current status and best practices of shared governance in US pharmacy programs

To characterize shared governance in US schools and colleges of pharmacy and recom-mend best practices to promote faculty engagement and satisfaction. Findings. The literature review revealed only one study on governance in a pharmacy school and some data from an AACP Faculty Survey. Of the 926 faculty members who responded to the survey, the majority were satisfied or very satisfied with faculty governance (64%) and the level of input into faculty governance (63%) at their school. Faculty members in administrative positions and those at public institutions were more satisfied with governance. The forum resulted in the development of five themes: establish a clear vision of governance in all areas; ensure that faculty members are aware of their roles and responsibilities within the governance structure; ensure faculty members are able to join committees of interest; recognize and reward faculty contributions to governance; and involve all full-time faculty members in governance, regardless of their tenure status.     Imaging of atherosclerosis, targeting LFA-1 on inflammatory cells with 111In-DANBIRT

    Background: 111In-DOTA-butylamino-NorBIRT (DANBIRT) is a novel radioligand which binds to Leukocyte Function-associated Antigen-1 (LFA-1), expressed on inflammatory cells. This study evaluated 111In-DANBIRT for the visualization of atherosclerotic plaque inflammation in mice. Methods and Results: ApoE−/− mice, fed an atherogenic diet up to 20 weeks (n = 10), were imaged by SPECT/CT 3 hours post injection of 111In-DANBIRT (~ 200 pmol, ~ 40 MBq). Focal spots of 111In-DANBIRT were visible in the aortic arch of all animals, with an average Target-to-Background Ratio (TBR) of 1.7 ± 0.5. In vivo imaging results were validated by ex vivo SPECT/CT imaging, with a TBR up to 11.5 (range 2.6 to 11.5). Plaques, identified by Oil Red O lipid-staining on excised arteries, co-localized with 111In-DANBIRT uptake as determined by ex vivo autoradiography. Subsequent histological processing and in vitro autoradiography confirmed 111In-DANBIRT uptake at plaque areas containing CD68 expressing macrophages and LFA-1 expressing inflammatory cells. Ex vivo incubation of a human carotid endarterectomy specimen with 111In-DANBIRT (~ 950 nmol, ~ 190 MBq) for 2 hours showed heterogeneous plaque uptake on SPECT/CT, after which immunohistochemical analysis demonstrated co-localization of 111In-DANBIRT uptake and CD68 and LFA-1 expressing cells. Conclusions: Our results indicate the potential of radiolabeled DANBIRT as a relevant imaging radioligand for non-invasive evaluation of atherosclerotic inflammation

    Force-velocity-power and Force-pCa Relationships of Human Soleus Fibers After 17 Days of Bed Rest

    Soleus muscle fibers from the rat display a reduction in peak power and Ca2+ sensitivity after hindlimb suspension. To examine human responses to non-weight bearing, we obtained soleus biopsies from eight adult men before and immediately after 17 days of bed rest (BR). Single chemically skinned fibers were mounted between a force transducer and a servo-controlled position motor and activated with maximal (isotonic properties) and/or submaximal (Ca2+ sensitivity) levels of free Ca2+. Gel electrophoresis indicated that all pre- and post-BR fibers expressed type I myosin heavy chain. Post-BR fibers obtained from one subject displayed increases in peak power and Ca2+ sensitivity. In contrast, post-BR fibers obtained from the seven remaining subjects showed an average 11% reduction in peak power (P \u3c 0.05), with each individual displaying a 7–27% reduction in this variable. Post-BR fibers from these subjects were smaller in diameter and produced 21% less force at the shortening velocity associated with peak power. However, the shortening velocity at peak power output was elevated 13% in the post-BR fibers, which partially compensated for their lower force. Post-BR fibers from these same seven subjects also displayed a reduced sensitivity to free Ca2+(P \u3c 0.05). These results indicate that the reduced functional capacity of human lower limb extensor muscles after BR may be in part caused by alterations in the cross-bridge mechanisms of contraction

    Inclusive V0V^0 Production Cross Sections from 920 GeV Fixed Target Proton-Nucleus Collisions

    Inclusive differential cross sections dσpA/dxFd\sigma_{pA}/dx_F and dσpA/dpt2d\sigma_{pA}/dp_t^2 for the production of \kzeros, \lambdazero, and \antilambda particles are measured at HERA in proton-induced reactions on C, Al, Ti, and W targets. The incident beam energy is 920 GeV, corresponding to s=41.6\sqrt {s} = 41.6 GeV in the proton-nucleon system. The ratios of differential cross sections \rklpa and \rllpa are measured to be 6.2±0.56.2\pm 0.5 and 0.66±0.070.66\pm 0.07, respectively, for \xf ≈−0.06\approx-0.06. No significant dependence upon the target material is observed. Within errors, the slopes of the transverse momentum distributions dσpA/dpt2d\sigma_{pA}/dp_t^2 also show no significant dependence upon the target material. The dependence of the extrapolated total cross sections σpA\sigma_{pA} on the atomic mass AA of the target material is discussed, and the deduced cross sections per nucleon σpN\sigma_{pN} are compared with results obtained at other energies.Comment: 17 pages, 7 figures, 5 table

    Paroxetine suppresses recombinant human P2X7 responses

    P2X7 receptor (P2X7) activity may link inflammation to depressive disorders. Genetic variants of human P2X7 have been linked with major depression and bipolar disorders, and the P2X7 knockout mouse has been shown to exhibit anti-depressive-like behaviour. P2X7 is an ATP-gated ion channel and is a major regulator of the pro-inflammatory cytokine interleukin 1β (IL-1β) secretion from monocytes and microglia. We hypothesised that antidepressants may elicit their mood enhancing effects in part via modulating P2X7 activity and reducing inflammatory responses. In this study, we determined whether common psychoactive drugs could affect recombinant and native human P2X7 responses in vitro. Common antidepressants demonstrated opposing effects on human P2X7-mediated responses; paroxetine inhibited while fluoxetine and clomipramine mildly potentiated ATP-induced dye uptake in HEK-293 cells stably expressing recombinant human P2X7. Paroxetine inhibited dye uptake mediated by human P2X7 in a concentration-dependent manner with an IC50 of 24 μM and significantly reduces ATP-induced inward currents. We confirmed that trifluoperazine hydrochloride suppressed human P2X7 responses (IC50 of 6.4 μM). Both paroxetine and trifluoperazine did not inhibit rodent P2X7 responses, and mutation of a known residue (F 95L) did not alter the effect of either drug, suggesting neither drug binds at this site. Finally, we demonstrate that P2X7-induced IL-1β secretion from lipopolysaccharide (LPS)-primed human CD14+ monocytes was suppressed with trifluoperazine and paroxetine

    Modeling crosshatch surface morphology in growing mismatched layers. Part II: Periodic boundary conditions and dislocation groups

    We present further developments and understanding of the commonly observed crosshatch surface morphology in strain-relaxed heteroepitaxial films. We have previously proposed that the crosshatch morphology is directly related with strain relaxation via threading dislocation glide which results in both surface step and misfit dislocation (MD) formation [see Andrews , J. Appl. Phys. 91, 1933 (2002)-now referred to as Part I]. In this article, we have used solutions for the stress fields and displacement fields for periodic MD arrays which include the effects of the free surface. These solutions avoid truncation errors associated with finite dislocation arrays that were used in Part I. We have calculated the surface height profile for relaxed films where the misfit dislocations were introduced randomly or the misfit dislocations were placed in groups with alternating sign of the normal component of their Burgers vector. We have calculated the surface height profiles where the slip step remains at the surface ["slip step only" (SSO)] and where the slip step is eliminated ["slip step eliminated" (SSE)] due to annihilation of opposite sense steps, such as could happen during growth or lateral mass transport. For relaxed films, we find that the surface height undulations, characteristic of crosshatch, increase with increasing film thickness for the SSO case, whereas the surface becomes flatter for the SSE case. Experiments on relaxed In0.25Ga0.75As films on (001) GaAs show that the surface height undulations in the [110] direction increase with increasing film thickness. Thus, we conclude that with increasing film thickness the crosshatch in the slow diffusion [110] direction is best described by the SSO case. (C) 2004 American Institute of Physics
